Syllabus of the lecture

Exposure to neurotoxic chemicals is suspected to play a role in the development and progression of neurological disorders. Neurotoxic chemicals are also known to have severe impacts on the ecosystem. Currently, testing for neurotoxicity is not required within the EU (REACH) as it is not clear how to assess neurotoxicity. Considering the increasing numbers of chemicals and the physiological and morphological complexity of the nervous system, it is a major challenge to test all substances for their neurotoxic potential, new advanced neurotoxicity assessment strategies need to be developed to fulfil these demands. During the last years we developed in several projects (DENAMIC, NeuroBox) novel (alternative) assessment strategies to detect neurotoxicity. Within the projects alternative model species like zebrafish (Danio Rerio) were applied and tested for their usability as screening tools. Novel Omics strategies were developed to measure metabolomics, proteomics and transcriptomic changes. Behavioral tests were performed after exposure to individual compounds and environmental mixture’s. Our results show that assessing neurotoxicity is complex and a tiered approach covering behavioral tests in combination with OMICS techniques seem to be a cost and time efficient way.

About the lecturer

Dr Jessica Legradi is a toxicologist working as Assistant Professor at the department Environment and Health at the VU in Amsterdam (Netherlands). At the moment she is also partially employed as researcher for the NeuroBox project at the Department of Ecosystem Analysis at the RWTH in Aachen (Germany). Her work is focused on the discovery of toxic mode of actions (MoA) of (developmental) toxic compounds. In the last years her main interest is on neurotoxic MoA. She worked successfully on several international and national projects. She is co-organizer of the Fish and amphibian embryos as alternative models in toxicology and teratology workshop and is chairing in the last 5 years the SETAC-Europe session Fish model species in environmental toxicology.
